Histology Solvents and Reagents
Introduction
J.T.Bakers?chemical know-how and its manufacturing and purification competencies form the basis of a comprehensive range of chemicals, stains and auxiliary products. Especially formulated and purified for use in the Histo-pathology. Together with our environmentally and human friendly clearing reagents, a range of solvents, alcohols, fixatives, stains and dyes are available through our network of international distributors.
Production
The histological reagents are produced using state-of-the-art production techniques in modern, certified manufacturing facilities, to assure a minimum of impurities. Production is centralized at J.T.Bakers?head-office in the town of Deventer, Holland.
J.T.Bakers?commitment to the International Responsible Care program is contributing to environmentally friendly and efficient manufacturing. The high quality of raw materials used guarantees the quality of the solutions produced.
The products are bottled under controlled conditions to prevent contamination and degradation, thus assuring maximum stability.
Certification and legislation
J.T.Bakers?procedures are all accredited to ISO-9001.
Also, in the near future, the J.T.Baker products will be produced and developed according the European IVD directive, 98/79/EC and will be carrying the CE Mark.
Quality Control
Every batch of the histological reagents and solvents is tested and released in accordance with ISO-9001 regulations. A highly educated, working in a well-equipped R&D and QC laboratory, guarantees lot-to-lot consistency and provides technical customer support.
Fixatives
Formaldehyde solutions
Effective and reproducible fixation requires a buffered Formaldehyde. Non-buffered Formaldehyde solutions are not consistent for its pH and when the solution becomes "acid" formation of Formaline-Heme pigments may appear. These pigments appear as black deposits in tissue.
Also the buffer concentration has proven to be an important factor.
J.T.Baker offers formaldehyde fixatives as well as a ready-to-use solution or as concentrated solutions which have to be diluted before use. All formaldehyde fixatives are phosphate buffered with an optimised buffer concentration.
Formalin Neutralizer
J.T.Baker recently developed a product which can help the users of formalin to discard their used or spilled formalin in a safe and easy way.
Discarding formalin in sewer systems can be dangerous for humans and is also extremely bad for the environment. A nice solution has been found in using Formalin Neutralizer, which is a powered chemical capable in neutralising formalin.
Formalin Neutralizer has a very easy working procedure and has been tested by a Sterlab Qualified environmentally laboratory

Bouins Fixative.
Bouins fixative is a picric acid based fixative used in a variety of general fixation procedures in Histopathology. Bouins solution is also a popular fixative for embryonic studies and skin, due to its excellent preservation of nuclei and chromosomes. Bouins is very compatible with the trichrome stains due to its mordanting effect on the tissue.
Cervix Spray Fixative
This is a specially designed non-toxic solution, to fix prepared cell smears in such a way that staining according to Papanicolaou gives optimised coloration of cell structures. Cervix Spray Fixative contains special glycols in an alcoholic medium. Cervix spray is supplied with an easy-to-use re-usable spray head
Environmentally and human friendly solvents to replace Xylene and Toluene.
UltraClear
UltraClear is an Iso-Paraffin based clearing reagent. Iso-paraffin’s are considered to be environmentally friendly and are therefore the perfect alternative for Xylene, Toluene and Citric clearing reagents.
UltraClear may be used as a Xylene replacement during tissue embedding and deparaffination processes.
UltraClear is non-toxic, non-carcinogenic, non-flammable, odourless, dermatological inert and easy to adapt with existing procedures.
UltraKitt?
Recently J.T.Baker developed a new mounting medium which is miscible with UltraClear, Xylene and Toluene. Some characteristics of UltraKitt are:
Contains no aromatic compounds or dangerous solvents
It is human and environmentally friendly
It flows out very nicely.
Examination within 1 hour.
Less migration of dyes.
No dry up effects of the slides.

Paraffin
UltraPar?54 - 56癈
UltraPar is a newly developed paraffin intended for tissue embedding and sectioning in histopathology. When melted UltraPar is a clear and bright solution. Special purification technology has resulted in particle free paraffin. This purity avoids damaging of expensive blades. UltraPar does not shrink and it does not crack during cooling. Penetration of UltraPar is classified as unique. Sectioning at 2 祄 is easily performed. The solubility of UltraPar in organic solvents and in UltraClear is extremely good.
Paraffin Cleaner
Laboratory equipment should remain in good condition and therefore regular cleaning is obliged. Microtomes are likely contaminated with paraffin residues. Paraffin Cleaner is a specially designed non-toxic, non-carcinogenic and non-flammable cleaning solution. It contains mineral oils in an alcoholic medium. Paraffin Cleaner is not suspected to cause OPS. In principle it may be considered as a non-harmful cleaning solution.
STAINING IN HISTOLOGY
Hematoxyline and Eosine Y ready-to-use solutions in HE staining techniques.
Hematoxyline-Eosine staining is the most preferred and used technique in routine and counter staining (regressive or progressive). The final results in terms of brightness and contrast obtained with this technique are superior over those obtained with many alternative methods. However, there are numerous qualities on the market manufactured at room temperature with an inconsistently solved cation (Aluminia). The presence of this cation is required as it acts like a mordant between the acidic nucleus and the stain.
Hematoxyline
J.T.Bakers?Hematoxyline, which is available according to Mayer or modified according to GillII/Harris, is a state of the art manufactured product. Modern technology allows a time and temperature controlled process. Continuous motion avoids precipitation and guarantees a final end product of extreme stability. This stability guarantees consistency, brightness and clearness.
Eosine Y and Scotch Buffer
J.T.Baker抯 Eosine Y is especially manufactured to match our Hematoxyline. Eosine Y is offered as an alcoholic or as an aqueous solution.The use of this Eosine and Scotch Buffer for coloring will guarantee consistent color development and will therefore contribute to extreme brightness and clearness of the stained tissue sections.

Papanicolaous Stains for optimized interpretation of cytology smears.
Cytopathology is the study of disease in cells. Despite all of the technological advances, which have occurred in medicine in the last 100 years, the Pap smear (named for its developer, Dr. George Papanicolaou) still is one of the few tests available to detect the presence of a premalignant lesion.
Experienced laboratory technicians classified the results obtained with the J.T.Baker PAP solutions, when used in combination with J.T.Baker Hematoxyline as counter stain, excellent and of superb color intensity. All stains used in the PAP staining solutions are purified and free from contamination.
J.T.Baker offers Papanicolaou 2A OG6, 2B Orange II and 3B EA 50 which are advised to be used in combination with Hematoxyline according to Mayer or with the Hematoxyline modified according to Gill II and Harris.

Hemacolour and May-Gr黱wald/Giemsa
Staining solutions for traditional methods such as May-Gr黱wald/Giemsa are offered but also and excellent fast alternative, Hemacolour is developed by J.T.Baker.
Hemacolour
Hemacolour was introduced because the need for fast and convenient staining methods has become more important. Hemacolour offers maximum correlation with the original reference method, excellent reproducibility and consistent staining within the shortest possible time.
Hemacolour is manufactured from certified stains and dyes and is available in various packsizes.
May-Gr黱wald/Giemsa
May Gr黱wald and Giemsa are recommended for use in traditional methods, such as staining of cytology specimen (mostly bone marrow and cervix tissue) and staining of histology specimen (mostly gastric sections). J.T.Baker offers May Gr黱wald and Giemsa with an optimized formulation so the colours are balanced. Also an optimized working procedure is available which helps when color tones need to be adapted.
Easy diluting (if needed) of the solutions can be done by using the J.T.Baker S鴕enson buffer solution.
Alcohols and Solvents
Alcohols
A broad range of ready-to-use alcohols and mixtures are available in convenient and easy-to- carry HDPE cans.
The quality of these alcohols matches your needs in Histo pathology and Cytology laboratory techniques. Extensive testing has proven the suitability and indicated excellent results when used in appropriate staining and tissue preparation procedures. The range of alcohols consists of Methanol, Ethanol, Ethanol denatured with Methanol, IPA and Benzyl alcohol.
Solvents
Alongside the widely used UltraClear, both Xylene and Toluene are available in a Histo grade quality. These solvents are especially prepared for use in all Histology technologies in the laboratory, like coverslipping and tissue embedding. These solvents are packed in tin cans with a special spout for easy pouring.
Special phosphate buffered saline solution
Immuno PBS
This solution is developed for use in immuno-histo-chemistry staining. The immuno PBS differs from the regular PBS in ionic strength, buffer composition and preservation. The buffer composition and ionic strength result in a much stronger antibody-antigen coupling. A much higher specificity is obtained, which contributes to a better diagnosis.
Also the preservative used is crucial. J.T.Baker uses an effective preservative, which optimises the colour development by the enzyme label.
The Immuno PBS is a 10-times concentrated solution, which should be diluted with distilled or deionised water prior to use.
New products
J.T.Baker continuously is expanding the product range. New products which are to be expected early 2001 are: Decalcifier, to decalcify bones, Papanicolaou EA 65 and Shorr reagent.
