Lipase LIPA 脂肪酶

这是常规双试剂的用法,B200ul提前加入,样品S
4ul在0秒加入,混合孵育200秒后加入C70ul,启动反应。
reagent triggered
表示用试剂触发反应,也就是样品加入后并不是反应的开始,需要再加一种试剂启动,这就是常说的双试剂R2加入后的启动反应。
这样空白读点区就应该是试剂和样品空白,也就是常见的R1+S的空白,所以在R2加入之前的区域,这里是200秒之前的黄色区域。初始反应区域是R2加入之后的1-5点之内绿色区域,红色区域才是反应读点。

这个LIP有个特殊性,就是先用A腔的330ul清洗反应杯,提前加入,冲洗过后,再加入B和C的试剂混合孵育长达300秒后再加入样品。可以看出试剂加入后也是呈速率上升反应趋势,所以采用的是速率2法,反应速率减去空白速率,也就是红色区域减去黄色区域。

胆固醇也是个特例,虽然用终点法2,但反应并没有达到终点。

高密也是个特例,选择的是终点法2,也就是终点反应减去空白反应。由于DXC没有固定点法和两点法选择,所以只能用终点法来替代固定点或两点法。
A先加入,0秒加入样品,200秒之前加入B,初始读点在B加入之后,空白读点并不是在B加入之前,而是在B加入之后的200秒以后,主读点在反应最后的终点区。这就是常说的两点终点法都在R2之后。这也是无奈之举,首先DXC除了终点和速率法没有其它方法,其次是B加入后明显的下降弯钩,将空白取在弯钩底部比取在B之前,更能准确的反应出反应变化的程度。

DXC-MC MC模块分为杯模块和ISE模块。 杯模块有最多六个,GLU、BUN是电极法,TP、ALB、Phos、CRE是光学法。 在杯模块中,系统检测一个样品的流程是40秒,在样品加入之前读取试剂空白,也就是参比读点,是黄色区域。样品加入之后读取反应读点,也就是红色区域。 样品反应结束之后,排空反应液,然后加入去离子水冲洗或直接接入新的试剂准备下一次检测。 搅拌存在整个过程,除了排空完毕加入去离子水或试剂的几秒钟停止外。 杯模块的校准是重复四次,去掉最大最小值进行计算。终点法的项目不显示Rate值,速率法的项目显示;delta ABS只在终点法的项目中显示;Init Read是初始读点,也就是空白读点。Final Read是最终读点。SET Point是校准靶值; SET Point unit是校准单位;Replicates Used表示保留下来哪两个重复值作为计算依据。 保留下来的重复值首先验证重复性,也就是背靠背。然后进行范围检查,超出设定的范围都会报告校准失败。 Albumin (ALBm)白蛋白 Endpoint (5 point average) 终点法反应,读点区有五个读点的值平均。校准选择的是1、3两个保留重复值。 Urea Nitrogen (BUNm)尿素氮 Rate (7 point moving medium) 速率法反应,7个点的值平均。 Creatinine (CREm)肌酐 Rate (5 point average) 速率反应,反应液排空后连续两次去离子水冲洗后才灌注试剂。读点开始前搅拌关闭,两次水洗时搅拌开启。 Glucose (GLUCm)葡萄糖 速率反应。 Phosphorus (PHOSm)磷 Rate (5 point moving average) 速率反应,反应读点前搅拌慢速运转,排空和水洗以及灌注试剂时高速运转。 Total Protein (TPm)总蛋白 Rate (least square linear regression) 速率法反应,
