【摘要】 目的探讨3种常见的影响全自动酶免分析系统检测结果的因素,提高EI ISA法检测结果的准确性。方法(1)随机选择对照组和实验组各44份无偿献血者血样,分别进行两种不同方式的抗凝,应用FAME全自动酶免分析系统进行HBsAg、抗一HCV、抗一HIV、TP 4个项目检测,比较两组检测结果。(2)在全自动加样仪的加样高度设置中,不断增加加样高度,观察FAME对4个质控血清平行孑L的检测效果。(3)在不同温度的FAME洗液下,对88份正常的无偿献血者血样进行4个项目检测,观察不同温度的洗液对检测结果的影响。结果 (1)除抗一HCV外,实验组中其他3项检测结果均有假阳性出现,且阴性标本的S/CO值明显高于对照组(P<O.001)。(2)当加样高度低于9.1 mm时,抗一HCV有假阴性出现,4项检测的CV值均较高,均一性差。(3)洗液温度低于l4℃时,除抗一HCV外,其他3项检测结果有假阳性出现,CV值较高,均一性差。结论标本的抗凝效果、全自动加样仪的加样高度和洗液温度对全自动酶免检测结果均有影响,只有在血样充分抗凝、加样高度和洗液温度适当等条件下,才能获得准确的检测结果。 【关键词】FAME全自动酶免系统 EI ISA 影响因素 【中图分类号】R446.1 R446.61 【文献标识码】A 【文章编号】l671—2587(2004)04—0254—03 Study of the influential factors on ELISA using the microlab FAME system ZHU Yangquan,X Changgen,DAI Ruidi, et a1.Zhenjiang Blood Center of Jiangsu Province,Zhenjiang 212001 [Abstract] Objective To study the three types of common factors which influence the results of the microlab FAM E system on EI ISA.M ethods (1)There were 44 random samples in experimental group and control group respectively,which were anti—curdled in different levels.Then FAME was used tO test HBsAg。anti—HCV,anti—HIV and TP respectively and the difference between the two groups were compared.(2)W hile increasing the dispensing height of the AT plus 2 in series on the basis of original default,the effects caused by them were assayed after using FAME through four items of parallel quality—controlled sera.(3)The results were detected using FAM E under the different temperature of wash solution,through 4 items of EI ISA on 88 samples of normal voluntary donors.Results (1)In the experimental group ,fault positive results emerged in three items except anti—HCV ,and the S f Co of negative samples was much higher than that i,I control group (P< 0.001).(2)W hen dispensing height was lower than 9.1 mm .fault positive results emerged in anti-HCV ,and the CV of four items were higher than norma1.(3)W hen the wash solution temperature was lower than l4℃ ,the fault positive results emerged and CV were higher than usual except anti—HCV.Conclusion The accurate resuits can be gained only when the blood samples were anti—curdled thoroughly,the dispensing height and the wash solution temperatures were suitable. 近年来,很多采供血机构相继引进了全自动酶免分析系统用于酶联免疫吸附试验(ELISA)检测,全自动酶免检测设备在过程处理的速度、准确性精密度和重复性方面均优于手工操作。本站于1999年引进FAME全自动酶免分析系统及配套AT plus 2全自动加样仪,在使用中发现,除了检测试剂本身的稳定性、灵敏度和特异性等因素影响检测结果外,很多操作方面的问题,如样本抗凝处理、加样高度设置及不同洗液温度等环节都会对检测结果的准确性造成不同程度的影响,甚至得到假阳性或假阴性结果,现报告如下。
